چکیده

1 2 The cell surface-associated extracellular adherence protein (Eap) mediates adherence of 3 Staphylococcus aureus to host extracellular matrix components, and inhibits inflammation, 4 wound healing and angiogenesis. A well-characterized collection of S. aureus and non5 S. aureus staphylococcal isolates (n = 813) was tested for the presence of the Eap-encoding 6 gene (eap) by PCR to investigate the eap gene as a specific diagnostic tool for identification 7 of S. aureus. Whereas all 597 S. aureus isolates were eap-positive, this gene was not 8 detectable in 216 non-S. aureus staphylococci comprising 47 different species and subspecies 9 of coagulase-negative staphylococci and non-S. aureus coagulase positive/variable 10 staphylococci. Furthermore, non-S. aureus isolates did not express Eap homologs, as verified 11 on transcriptional and protein levels. Based on these data, the sensitivity and specificity of the 12 newly developed PCR targeting the eap gene were both 100 %. Thus, the unique occurrence 13 of Eap in S. aureus offers a promising tool particularly suitable for molecular diagnostics of 14 this pathogen. 15 AC CE PT ED on N ovem er 3, 2017 by gest ht://jcm .sm .rg/ D ow nladed fom

Background: Staphylococcus aureus and Staphylococcus epidermidis are major human pathogens of increasing importance due to the spread of antibiotic resistance. Novel potential targets for therapeutic antibodies are products of staphylococcal genes expressed during human infection. Previously, the secreted and surface-exposed proteins among seroreactive antigens have been discovered. Furthermore...

Identification of Toxic Shock Syndrome Toxin-1 (TSST-1) gene in Staphylococcus aureus isolated from bovine mastitis milk

Staphylococcus aureus is a major causative pathogen of clinical and subclinical mastitis of dairy domestic ruminants. This agent produces a variety of extracellular toxins and virulence factors including Toxic Shock Syndrome Toxin-1 (TSST-1) which is the major cause of Toxic Shock Syndrome (TSS). In this study 58 S. aureus isolates obtained from 9 dairy herds in East and West Azerbaijan provinc...

Detection of eap Gene in Clinical Isolates of Staphylococcus aureus Before and After In vitro X-Irradiation and its Association with Antimicrobial Susceptibility

Extracellular adherence protein (Eap) constitutes an important virulence factor enhancing binding and internalization of Staphylococcus aureus into eukaryotic cells. In the present study 63 S. aureus isolates from cancer patients were examined for the existence of the Eap-encoding gene (eap) before and after in vitro X-irradiation using PCR amplification to determine whether this gene could be ...

Enterotoxin gene profiles among Staphylococcus aureus isolated from raw milk

Milk is considered a nutritious food because it contains several important nutrients including proteins and vitamins. Conversely, it can be a vehicle for several pathogenic bacteria such as Staphylococcus aureus. This study aimed to analyze the frequency of genes encoding the nine Staphylococcal enterotoxins (SEs) and enterotoxin gene profiles in S. aureus isolates derived from raw bovine milk....

Antibiotic Resistance and Detection of femA Gene in Staphylococcus Aureus Isolates from Raw Milk

ABSTRACT         Background and Objective: Antibiotic-resistant Staphylococcus aureus strains have become a problem in treatment of infections caused by S. aureus. This study aimed to evaluate antibiotic resistance in S. aureus isolates from raw milk and detect femA gene in these isolates, as a confirmatory test for identification of S. aureus species. ...
